Indications for treatment of Imbruvica
Imbruvica is an anti-cancer drug that contains the active substance ibrutinib. It belongs to a class of drugs called protein kinase inhibitors.
Imbruvica is used to treat the following blood cancers in adults:
- Neuroblastoma lymphoma (MCL): a type of cancer that affects lymph nodes, when the disease returns or does not respond to treatment.
- Chronic lymphocytic leukemia (CLL) / Small lymphocytic lymphoma (SLL): a type of cancer that affects white blood cells called lymphocytes is also related to lymph nodes. Imbruvica is used in patients who have not previously been treated for CLL / SLL or have received at least one previous therapy.
- Waldenstrom macroglobulinaemia (WM): a cancer that affects white blood cells called lymphocytes. It is used when the disease has returned or does not respond to treatment or in the first treatment for patients who are not suitable for chemotherapy immunotherapy.
- Regional lymphoma (MZL) requires oral or injection (systemic) therapy and has received a previous treatment.
- Chronic graft versus host (cGVHD): after failure of 1 or more lines of systemic therapy
The mechanism of action of Ibrutinib
- The active ingredient in Imbruvica is ibrutinib that is active against carcinogenic B lymphocytes, a type of white blood cell affected by these diseases.
- It does this by blocking an enzyme called Bruton’s tyrosine kinase (Btk), which promotes the survival of B lymphocytes and their movement to the organs where these cells normally divide. By blocking Btk, ibrutinib reduces B lymphocyte survival and migration thus slowing the progression of cancer.
Contraindicated
- Are allergic to any of the ingredients of Imbruvica.
- Children under 18 years of age do not recommend this medication.
- Pregnant and lactating women.

Dosage and usage of Imbruvica
Dosage recommended
MCL and MZI
- The recommended dose of Imbruvica is 560mg (4 capsules) once daily.
CLL / SLL, WM and cGVHD
- The recommended dose of Imbruvica is 420mg (3 capsules) once daily.
Using
- Take capsules by mouth with a glass of water.
- Use the drug at the same time every day.
- Swallow whole capsules, do not break or chew them.
- Do not stop taking this medication unless your doctor tells you to
Warnings and precautions when using Imbruvica
- Hemorrhage: Problems with bleeding are common during Imbruvica treatment and can also be serious and can lead to death.
- Infection: may occur during treatment with Imbruvica, can be serious and can lead to death.
- Decreased blood cell count: Reduced blood counts (white blood cells, platelets and red blood cells) are common with Imbruvica.
- Arrhythmia: People with a history of heart disease using Imbruvica will cause serious heart rhythm problems that can lead to death.
- Hypertension: Imbruvica treatment can increase hypertension and worsen the condition in patients with a history of hypertension.
- The second malignant tumors: using Imbruvica can develop new cancer in other parts of the body.
- Tumor lysis syndrome (TLS): TLS is caused by the rapid disruption of cancer cells. TLS can cause kidney failure, abnormal heart rhythms, seizures and sometimes death.
- Embryotoxicity : Imbruvica may harm an unborn baby. Pregnant women should not use this medication.
Side effects of Imbruvica
The most common side effects of Imbruvica in adults with B cell malignancies (MCL, CLL / SLL, WM and MZL) include:
- Diarrhea
- Nausea
- Muscle and bone pain
- Tired
- Rash
- Fever
- Bruising
The most common side effects of Imbruvica in adults with cGVHD include:
- Tired
- Bruising
- Diarrhea
- Mouth sores
- Muscle spasms
- Nausea
Diarrhea is a common side effect in Imbruvica users. Drink plenty of water during treatment to help reduce the risk of excessive dehydration from diarrhea. Tell your doctor if you have diarrhea that does not go away.

Other drug interactions with Imbruvica

- Blood thinners such as warfarin, heparin or other medications for blood clots
- Supplements may increase the risk of bleeding like fish oil, vitamin E or flaxseed.
Interact with food
Do not take Imbruvica with grapefruit or Seville oranges (bitter oranges), which may increase the amount of Imbruvica in the blood, which is associated with a higher risk of side effects.
