What is Afatinib?
Afatinib 40mg is a medicine used to treat a type of lung cancer called non-small cell lung cancer. It is used especially in adults with advanced cancer in the following situations:
- when the cancer has a genetic mutation for a protein called EGFR and has not previously been treated with tyrosine kinase inhibitors.
- when the cancer is a squamous cell type (from cells of the pleura) and has worsened despite platinum-based chemotherapy.

- Giotrif has been shown to significantly slow disease progression in non-small cell lung cancer patients.
- In a main study of 345 patients with tumors with the EGFR mutation gene, the Giotrif-treated patients lived an average of 11 months with no worse disease than 7 months for patients treated with two types. Other cancer medications are pemetrexed and cisplatin.
- In a second study of 795 patients with squamous cell carcinoma patients, Giotrif-treated patients lived an average of 2.6 months, but the disease was not worse than 1.9 months for treated patients. Another cancer drug is erlotinib.
The mechanism of action of Giotrif
- The active substance in Giotrif, afatinib, is an ErbB family blocker. This means it blocks the action of a group of proteins called ‘ErbB families’ found on the surface of cancer cells and is involved in stimulating cell division.
- By blocking these proteins, afatinib helps control cell division and thus slows the growth and spread of non-small cell lung cancer.
- The EGFR protein is part of the ErbB family. Mutated lung cancer cells with the EGFR protein are particularly sensitive to afatinib.

- Use Giotrif exactly as your doctor told you to use it.
- Your doctor will tell you how much Giotrif to take and when to take it. Do not change your dose or stop Giotrif unless your doctor tells you to.
- Take Giotrif on an empty stomach at least 1 hour before a meal or 2 hours after a meal.
- If you miss a dose of Giotrif, take it as soon as you remember. If within 12 hours of your next dose, skip the dose and only take the next dose at regular intervals.
